Luke 1:35 speaks of the holiness of Mary and the child. Something to be holy must be uncontaminated (Lev 12:5-7, Eze 16:6-9). The boy born is uncontaminated by any blood elements. Here mention is made about the physical aspect of the virginity of Mary. The Protogospel of James dated around 150 AD, affirms the virginity of Mary at the Birth of Jesus.
